Description
Offer Description: The Rector of Wrocław University of Environmental and Life Sciences announces an open competition for the position in the research-technical group (F/M/X) in the project entitled “Personalized antibody-drug conjugates activated by matrix metalloproteinases for solid tumors”, acronym: PACMAN, Virtual Research Institute (WIB); managing entity: Łukasiewicz Research Network – PORT; implemented at the Department of Pharmacology and Toxicology Position title: Research-Technical Employee (Technical support) Field: Veterinary Sciences Discipline: Veterinary Science Location: Wrocław Contract type: Employment contract ( umowa o pracę ) Working time: 1/2 Part-time (0.5 FTE) Organisational unit: Department of Pharmacology and Toxicology, Laboratory of Preclinical Research, Faculty of Veterinary Medicine, Wrocław University of Environmental and Life Sciences The PACMAN project develops a new class of antibody–drug conjugates (ADCs) that do not require internalisation and are selectively activated extracellularly by matrix metalloproteinases (MMP‑2, MMP‑9 and MMP‑14) in the microenvironment of solid tumours. Position description: Specialist for the derivation and characterisation of primary colorectal cancer cell lines and their application in the assessment of the cytotoxicity of ADC conjugates. Main tasks and responsibilities: derivation of primary colorectal (colon) cancer cell lines from tissues obtained from patients undergoing surgical resection of primary tumours and metastatic lesions; optimisation of protocols for the isolation and culture of primary and subsequent cell lines to obtain viable cancer cultures demonstrating high proliferative activity while maintaining the stability of the cellular phenotype, monitored using cytometric analyses; preparation of a cell collection, including characterisation of cellular profiles based on parameters associated with proliferative activity (PDT, clonogenicity), metabolic activity (MTS), secretory activity (ELISA) and transcriptional activity (RNA-Seq/RT-qPCR); interpretation of tissue and cellular context through comparison of patient-derived material with the properties of primary cell lines, taking into account preservation of the tumour phenotype, tumour heterogeneity, marker expression and the limitations of the in vitro model; analysis of the cytotoxicity of ADC conjugates against primary colorectal cancer cell lines using cytotoxicity assays and methods enabling monitoring of the molecular status of cells; maintenance of accurate, comprehensive and standardised research documentation, including raw data, experimental conditions, procedural records, analysis of results and data archiving in accordance with the principles of good scientific practice. Requirements: higher education qualifications confirmed by at least a Master's degree in biological, biomedical, medical, pharmaceutical, veterinary or related sciences; possession of a doctoral degree in the above areas will be considered an additional advantage; experience in conducting preclinical research using in vitro and ex vivo models; experience with techniques used in cell culture, including the derivation and characterisation of primary cell lines, documented by publications; experience in assessing compound cytotoxicity using high-throughput cellular assays, documented by publications; experience in molecular biology techniques, documented by publications; commitment to comply with the non-compete rules in accordance with the WIB Programme guidelines. Selection criteria: The candidate’s scientific and professional experience, as well as competences to perform tasks within the research project. The scope of previous research activities and their alignment with the subject of the Research Task. The quality and scope of the candidate’s previous scientific achievements, in particular scientific publications, participation in research projects, awards and distinctions received, and references issued by independen
