PROJECT SUMMARY/ABSTRACT
Given that internalizing disorders are highly prevalent starting in childhood, understanding early risk factors for
these symptoms is crucial for mitigating long-term negative effects on academic performance, social
functioning, and overall quality of life. Two critical early risk factors—irritability and negative life events—are
strongly linked to the development of internalizing disorders. Irritability is multifaceted, with both tonic (grouchy
mood) and phasic (temper outburst) components. However, little is known regarding whether differential
longitudinal interactions among phasic and tonic irritability and negative life events influence internalizing
symptoms. Additionally, the neural mechanisms underlying these processes remain unknown. Since irritability
is one of the primary reasons that children are brought in for psychiatric care, it is critical to understand if
irritability’s two core facets (tonic and phasic) predict divergent symptom trajectories and to deepen our
knowledge regarding the underlying pathophysiology that may influence these differences. Longitudinal work is
thus needed to examine the distinct impact of phasic and tonic irritability on internalizing symptom trajectories
in the context of negative life events, and to investigate associated neural substrates. This proposal will utilize
data from the Adolescent Brain Cognitive Development (ABCD) Study, a longitudinal sample of over 11,000
youth followed from 9-15 years, with resting-state fMRI completed at two-year intervals, and measures of
irritability, negative life events, and internalizing symptoms collected annually. Using linear mixed effect
modeling, this study will examine the interactive effects of phasic and tonic irritability with negative life events
on internalizing symptom trajectories (Aim 1). This project will also investigate the relationship between phasic
and tonic irritability and network connectivity in the context of high negative life events over time, using
longitudinal canonical correlation analysis (CCA) (Aim 2). This predoctoral fellowship award will bring together
a team of experts in their respective fields and allow for the PI Archer to receive advanced training in
developmental neuroscience, resting-state fMRI network connectivity analysis, and advanced quantitative
methods. Together, the research project and training will provide the fellow with the opportunity to develop
necessary skills for a productive career as a clinical scientist investigating how irritability’s adaptive and
maladaptive features impact internalizing symptoms, with the hopes of informing the development of targeted
and personalized interventions.
