PROJECT SUMMARY
The goal of this project is to establish and leverage a new paradigm to optically measure and track synaptic
weights in the brains of awake animals as they learn and store information. By combining cellular resolution
optogenetic photostimulation and a genetically encoded voltage indicator for subthreshold voltage imaging –
we will develop a system with entirely new capabilities that will be broadly beneficial for neuroscience research
and highly impactful for circuit and systems neuroscience. The system will combine high throughput single cell
photo-stimulation with subthreshold postsynaptic membrane voltage recording to map and monitor and map
synaptic weights across thousands of candidate synaptic connections. We intend to capitalize on this new
platform to map and track synaptic weights in the visual cortex while an animal learns a visual perceptual task
and during deprivation induced plasticity. In so doing, we aim to directly reveal the biological learning
algorithms for experience dependent learning. Unlike all prior approaches in mammals which could only
indirectly relate changes in neural activity to changes in synaptic connections with learning, our new approach
will allow us to directly reveal how learning dynamically shapes synaptic weights in a neural network to give
rise to altered neural representations that implement learning.
