SUMMARY
Most common diseases are caused by both genetic factors and environmental factors. Gene-Environment
interactions (GxE) describe when genetic variation influences how individuals respond to their
environments. One set of genes that is particularly important to environmental response is the Aryl
Hydrocarbon Receptor (AHR) and associated genes that are involved in metabolizing foreign chemicals in
the body. We refer to these 29 genes as the AHR Pathway. In this research project, we will show how
genetic variation in these genes interacts with common environmental exposures such as diesel exhaust
to influence human diseases, such as coronary artery disease. To test this hypothesis, we will use the
Personalized Environment and Genes Study (PEGS), which is a human cohort study designed specifically
for identifying GxE. We map GxE and Polygenic Risk Score x Environment interactions in the PEGS cohort.
